Adanın sahəsi 41 214 km²[1] təşkil edir. Adaların dünyada sahəsinə görə sıralamasında 34-cü yerdə gedir. Sahil xətdinin uzunluğu 1944 km[2] təşkil edir.
Sauthempton adası Melvill yarımadasından Frozen-Streyt boğazı vastəsi ilə ayrılır[3]. Hudzon körfəzinin qərb sahilindən isə onu Ros-Velyam boğazı ayırır. Ən hündür nöqtəsi Matiasen dağıdır. Bell yarımadası cənub-şərqdə yerləşir[4].
Sauthempton adası Kanada ərazisində qış vaxtına keçməyən az saylı ərazilərdəndir.

Adada yeganə yaşayış məntəqəsi Koral-Harbordur. 2001-ci il məlumatına əsasən qəsəbədə 721 insan yaşayır.
